Purified KIF3B was injected into rabbits four times and the rabbits were bled. The serum was then affinity purified us- ing a KIF3B-bound activated Thiol-Sepharose 4B affinity column (Phar- macia Inc.). Western blotting of the final antibodies was carried out to confirm no cross-reaction with KIF3A. For the motility inhibition assay, purified antibodies were dialyzed against a PEM buffer (0.1 M Pipes, pH 6.8, 1mM EGTA, i mM MgC12)."

Overex- pressed KIF3A protein was loaded onto SDS-PAGE and the band corre- sponding to KIF3A protein was cut out of the preparative gel. 500/~g of excised KIF3A protein was mixed with complete Freund's adjuvant and in- jected into the axial lymph nodes of two rabbits. The rabbits received three booster shots with 250 #g of excised KIF3A mixed with incomplete Freund's adjuvant at 14-d intervals. On the third day after the third booster, the rabbits were bled from the ear vein and tested by immunoblotting analysis. Subse- quently, this serum was affinity purified using a KIF3A-bound triopropyl- Sepharose 6B affinity column."
